What "dsip peptide" names, and what it does not
DSIP is a nine-residue synthetic peptide, Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, written WAGGDASGE; formula C35H48N10O15, average mass 848.8 Da, PubChem CID 68816. The 1977 paper cited at the foot of this page reported it from rabbit blood and named it for an electroencephalogram observation, and the 2006 review cited beside it is titled, accurately, a still unresolved riddle: no receptor has been identified and the mechanism was never settled. Its nonproprietary name is emideltide, which is how the FDA lists it. Listings write DSIP, DSIP peptide and delta sleep-inducing peptide; one molecule.
The name describes an early observation, not a demonstrated mechanism, and the most useful thing a buyer can know about DSIP is that the research is thin and old; this page makes no claim about what the peptide does. What it can say precisely is what the peptide is: a short, acidic sequence with one tryptophan, two acid side chains and a free C-terminus, cheap to make, and easy to get wrong in three specific ways that a certificate can show.
The three ways are a missing glycine (there are three, and a deletion runs close to the main peak), an oxidised tryptophan (the indole ring picks up oxygen on light exposure, adding 16 or 32 Da), and the salt. With two acids and a free terminus the peptide holds counter-ion tightly, and a trifluoroacetate lot can be a fifth salt by weight, so the net content line decides what a labelled milligram means.
The reference point for identity is public. PubChem lists Delta sleep-inducing peptide as CID 68816 with molecular formula C35H48N10O15, an average molecular weight of 848.8 Da and a monoisotopic mass of 848.33 Da. Those are the numbers a mass spectrometer is checked against, and the number a certificate must print beside the observed mass.
In plain terms: Delta sleep-inducing peptide, a man-made peptide nine building blocks long. No approved product, no confirmed receptor, and a name that describes an early observation rather than a demonstrated mechanism. The research is thin and old, and that is the most important thing to know about it.

The price signal is the salt and the light. The certificate a DSIP lot must carry
Three tests answer three different questions, and a certificate of analysis for DSIP is only complete when it carries all three for the exact lot on the vial. Identity is settled by mass spectrometry, and the number to look for on a DSIP certificate is 848.33 Da monoisotopic (or 848.8 Da average, if the laboratory reports the deconvoluted average mass). The observed mass should sit within the instrument's stated tolerance of that figure, and the certificate should print both the theoretical and the observed value, not just a tick.
Purity is a separate question, answered by HPLC: how much of what came off the column was the compound. The figure is only interpretable with its method beside it (column, gradient, detection wavelength) and a date, which is what ICH Q2(R2) means when it asks that an analytical procedure be shown fit for its purpose. A percentage with no method and no date is a number somebody typed, and a certificate that carries every test but for a different lot number than the vial is the most common failure of all and the easiest to catch. Net content is the third test and the one most often missing: the milligrams of DSIP actually in the vial, measured, against the size printed on the label.
Identity is the observed mass against 848.330 Da monoisotopic (848.8 Da average), seen in electrospray as m/z 849.3 for the singly charged ion or 847.3 in negative mode, which suits an acidic peptide; the certificate should print theoretical and observed. A mass 57 Da low is a missing glycine, a common deletion in a sequence with three of them; peaks at +16 and +32 Da are oxidised tryptophan, honest at a fraction of a percent and a warning as a main peak. A certificate reading "conforms" with no number has not distinguished DSIP from its own deletion product.
Purity by HPLC should show the chromatogram with its column, gradient and wavelength, since the des-glycine deletion elutes near the parent on a generic method and the oxidised forms elute earlier; the tryptophan gives DSIP a strong absorbance at 280 nm, which is a convenient second wavelength for confirming the main peak is the tryptophan-containing molecule. Net content is the line that pays on a small acidic peptide: quantitative HPLC against a standard or amino acid analysis tells you how many of the labelled milligrams are peptide rather than trifluoroacetate or acetate and water, and the salt belongs beside that figure.
Where to buy DSIP: five checks before you order from anyone
Every seller of dsip peptide says "third-party tested". The phrase costs nothing. These five checks take about four minutes per listing and separate a tested lot from a tested-sounding one.
- The lot on the vial is the lot on the certificate. Read the printed lot number, then open the certificate and match it. A certificate for "DSIP" with no lot, or a lot that does not match, describes some other vial.
- The laboratory is named and has its own lookup. The report should come from a laboratory the seller does not own, with a search code you can enter on the laboratory's own site. LabFirst lots are tested by Freedom Diagnostics and looked up by accession number; anything that can only be verified on the seller's own page has not been verified.
- The theoretical mass is printed, and it is the right one. For DSIP that is 848.33 Da monoisotopic; a certificate that prints a different figure has either measured a different molecule or copied a template.
- Purity has a method and a date. An HPLC percentage should name the method and the run date. A figure that reads "99%+" with neither, or one date reused across every product a seller lists, is decoration.
- Net content is stated in milligrams. The label size is a claim; the certificate's net content is the measurement. If the certificate has no net content line, the purity figure describes the powder, not the vial, and the price per mg you calculated is fiction.
How listings for DSIP specifically mislead:
- Copy that presents the name as a finding. DSIP was named for an observation in 1977, no receptor has been identified, and a certificate describes a vial, not a mechanism.
- "DSIP 99%" with no observed mass. Three glycines make a des-glycine deletion likely and it runs close to the parent; a purity number without the mass cannot show which peptide the main peak is.
- A 10 mg label with no salt form and no net content. Two acid side chains and a free terminus hold counter-ion tightly; a trifluoroacetate lot can be a fifth salt by weight.
- "Pharmaceutical grade DSIP". There is no USP monograph and no approved product; the FDA placed emideltide (DSIP) in Category 2 of its 503A list in September 2023, so no pharmacy may compound it.

Tryptophan is the storage story. The indole ring oxidises on exposure to light and air, slowly in the sealed dry cake at 2 to 8 C in the dark and quickly in a solution left on a lit bench, so a prepared DSIP solution is kept cold and dark, dated, and used inside the laboratory's own stability record rather than any figure on a label. The aspartate at position 5 can slowly rearrange in solution through a cyclic imide, a reaction that depends on pH. The dry cake's other enemy is water drawn from humid air once the vial is opened, which an acidic peptide salt takes up readily; open a vial once, for its purpose, and never re-seal one for storage.

Regulatory status, and what this page is not
No approved medicine contains DSIP and no USP or Ph. Eur. monograph defines an acceptable lot. In September 2023 the FDA placed Emideltide (DSIP) in Category 2 of its interim list of bulk drug substances nominated for compounding under section 503A, the category for substances it has identified as raising significant safety risks, which bars licensed pharmacies from compounding with it. That listing concerns pharmacies, not laboratory material, and is quoted because "pharmacy grade" appears in listings for a substance no pharmacy may currently compound.
