TB-500 is a piece of thymosin beta-4, so the thymosin beta-4 research is where its mechanism comes from. That protein grabs single actin units one at a time and holds them out of play, which stops actin filaments growing and shifts the balance between loose actin and built-up actin. Crystal images showed how it does it: a short section called the WH2 motif caps both ends of an actin unit, and the hand-off between thymosin beta-4 and another protein, profilin, works because their grip sites overlap slightly.
Since building actin is what moves a cell, anything that changes how much free actin is available changes how cells move, and that is the thread running through the tissue-repair studies.
