And at the GLP-1 receptor it does not simply act weaker, it acts differently, favoring one signalling route (cAMP) over another (beta-arrestin) and pulling the receptor inside the cell less than the body’s own GLP-1 does. That matters because beta-arrestin1 appears to hold back the insulin response to GLP-1 but not to GIP or to tirzepatide, so the lopsided signalling is offered as part of how the molecule works rather than a footnote. At the GIP receptor it behaves much like the body’s own GIP.

The fatty acid lets it ride on albumin in the blood, which is how it stays in circulation for so long, the same trick used across this class.
