What is FOXO4-DRI?
FOXO4-DRI is also listed and searched as FOXO4-DRI peptide. They all mean the same compound. Supplier lists write it as peptide FOXO4-DRI as well; every one of those is this record. A name is not a test, which is why the lot record further down counts for more than the wording on a label.
Chemically: molecular formula C228H388N86O64, average mass 5358 g/mol (PubChem CID 167312269). Those figures are what the identity test on a certificate is read against.
A D-retro-inverso peptide: a cell-penetrating peptide built from D-amino acids in reversed sequence order, designed to resist proteolysis while preserving the side-chain topology of the native FOXO4 sequence.
Perturbs the interaction between FOXO4 and p53. In senescent cells this causes p53 nuclear exclusion and cell-intrinsic apoptosis — a senolytic mechanism that exploits how senescent cells specifically avoid dying.
Senescent cell clearance, doxorubicin-induced chemotoxicity, and fast-ageing and naturally aged mouse models.
No human data, no approved product. The published in vivo work is explicitly qualified as being conducted under conditions where the peptide was well tolerated, which is itself a statement about a therapeutic window.

Where to buy FOXO4-DRI: what to check before you order from anyone
FOXO4-DRI for sale, or FOXO4-DRI peptide for sale, means this page: one record, 2 sizes. You can buy FOXO4-DRI online here with guest checkout; verified in-stock orders placed before 12 PM Pacific are dispatched the same day.
Before ordering FOXO4-DRI from anyone, us included, run the 5 checks on verifying a certificate; the first is that the report resolves on the laboratory's own site, and a seller who cannot pass them is asking for the trust a certificate exists to replace.

The FOXO4-DRI certificate: lot, purity, identity, and how to verify it
A certificate answers identity, purity and net content as three separate questions, each with the method that produced it and the date it was run, how to read one covers what each field does and does not cover. For FOXO4-DRI that means a mass spectrum landing on 5,358 g/mol for C228H388N86O64, the figure published against PubChem CID 167312269.
FOXO4-DRI certificates post here and in the certificate index when Kovera Labs reports on the lot.

FOXO4-DRI research: what the published studies examine
Status: No FDA-approved application. This compound is sold as laboratory material. Where research on it has been published, it sits at the animal, cell or early-human stage, and this record does not summarize what it found. A supplier is not a neutral judge of evidence about its own catalog.
FOXO4-DRI is a designed peptide with two distinct pieces of engineering behind it. The mechanism is a targeted disruption of a protein-protein interaction: senescent cells accumulate irreparable damage but avoid apoptosis, and FOXO4 was identified as a pivot in how they manage that, holding p53 in a configuration that keeps the cell alive. A peptide that perturbs the FOXO4-p53 interaction causes p53 nuclear exclusion and cell-intrinsic apoptosis selectively in senescent cells — the selectivity comes from the fact that non-senescent cells are not depending on that interaction for survival.
The second piece of engineering is the D-retro-inverso construction: the peptide is built from D-amino acids in reverse sequence order, which preserves the spatial arrangement of side chains that determines binding while making the backbone unrecognisable to proteases. That is a real and elegant stability solution, and it also means the molecule is a mirror-image construct rather than a natural sequence, with the analytical implications that carries.
| Reported observation | Model | Source |
|---|---|---|
| FOXO4 was identified as a pivot in senescent cell viability. A FOXO4 peptide perturbing the FOXO4-p53 interaction caused selective p53 nuclear exclusion and cell-intrinsic apoptosis in senescent cells. Under conditions where it was well tolerated in vivo, the peptide neutralized doxorubicin-induced chemotoxicity, and restored fitness, fur density and renal function in both fast-ageing Xpd mice and naturally aged mice. | human and mouse cell lines, fast-ageing Xpd mutant mice and naturally aged mice | Baar and colleagues, Targeted apoptosis of senescent cells restores tissue homeostasis in response to chemotoxicity and aging, Cell (2017)Erasmus MC with the Buck Institute. Note the authors' own qualifier: "under conditions where it was well tolerated" — the tolerated window is doing real work in that sentence. |
Open questions. This is a high-profile single paper in a major journal, and the qualifier the authors themselves attach deserves more attention than it usually gets: the in vivo benefits were observed under conditions where the peptide was well tolerated, which is an explicit acknowledgement that a tolerability boundary exists and that the useful window sits below it. The mechanism is deliberate induction of apoptosis, so the compound is cytotoxic by design and its selectivity depends entirely on senescent cells' particular dependence on the FOXO4-p53 interaction.
Senescent cells are not uniformly harmful either — they have roles in wound healing and tumour suppression — so clearing them is not straightforwardly beneficial. There are no human data, no approved product, and the D-retro-inverso construction means an ordinary peptide identity assay must confirm stereochemistry, not just mass and sequence, which is a harder analytical question than it appears.
Each row above describes an observation in the stated model. None of it describes what FOXO4-DRI would do in a person or an animal outside that model, and none of it is a recommendation to use it for anything.
How to store FOXO4-DRI: refrigeration and shelf life
Keep the sealed vial cold and out of the light and it stays as it left the lab. Once it is mixed into a solution, how long it holds up depends on the compound, what it was mixed with, how strong the solution is and how cold it is kept — too many variables for anyone to print one shelf life on a label. So laboratories write the date on whatever they prepare and keep their own record of it.
For solution preparation arithmetic on the bench, concentration, per-graduation values and molarity, use the vial concentration calculator, which opens with the FOXO4-DRI sizes stocked here already loaded. It covers laboratory measurement only and contains no administration guidance.
