PTD-DBM carries the Dishevelled-binding motif itself, so it competes with CXXC5 for that interaction; blocking the brake activates the pathway.

Open questions. Read the in vivo result carefully: the accelerated wound healing in mice was produced by PTD-DBM together with valproic acid, and the authors describe the combination as synergistic. The peptide's own contribution separate from the GSK3-beta inhibitor is not established by that experiment, and summaries of this compound routinely drop the second agent. Beyond that, the work is from a single laboratory, there are no human data and no approved product.
One mechanistic caution deserves stating: Wnt/beta-catenin is a fundamental developmental and proliferative pathway active well beyond skin, and pharmacologically removing one of its negative regulators is a blunt intervention — sustained pathway activation is implicated in tumorigenesis in several tissues, which is exactly why the therapeutic interest here is in topical and local application rather than systemic.
