In short

ACE-031 is fusion protein of the activin receptor type IIB extracellular domain with an IgG1 Fc region — a soluble receptor decoy, not a peptide. ACE-031 is sold here for laboratory research in 1 size (1mg), from $60.00 a vial. It is not an FDA-approved medicine. Research use only, and never for people or animals.

What is ACE-031?

ACE-031 is also listed and searched as ACE-031 peptide, ramatercept. They all mean the same compound. Supplier lists write it as peptide ACE-031 as well; every one of those is this record. A name is not a test, which is why the lot record further down counts for more than the wording on a label.

Class

Fusion protein of the activin receptor type IIB extracellular domain with an IgG1 Fc region — a soluble receptor decoy, not a peptide.

Mechanism

Binds myostatin and related TGF-beta superfamily ligands in circulation, preventing them from reaching activin type II receptors and thereby removing their inhibitory signal on muscle.

Where it appears in the literature

Developed by Acceleron Pharma and tested in a randomised, double-blind, placebo-controlled ascending-dose trial in ambulatory boys with Duchenne muscular dystrophy.

What is not established

No approved product. The clinical trial was stopped early over safety findings, and the efficacy trends reported were not statistically significant.

ACE-031 for sale: 1mg vials, price per mg

ACE-031 is sold in 1 size here, 1mg, from $60.00 a vial. Every way of buying is in the table; the per-mg figure under each price is the one that compares across sizes.

Volume tiers are per size and do not combine across sizes (see bulk pricing). Subscribe & Save takes 6% every month, 4% every 2 months or 2% every 3 months off from shipment 2, two shipments minimum, and stacks on the volume tier: the lowest this record ever bills is $47.94 per mg, twenty 1mg vials on a monthly subscription from shipment 2 onward.

Before ordering ACE-031 from anyone, us included, run the 5 checks on verifying a certificate; the first is that the report resolves on the laboratory's own site, and a seller who cannot pass them is asking for the trust a certificate exists to replace.

The ACE-031 certificate: lot, purity, identity, and how to verify it

A certificate answers identity, purity and net content as three separate questions, each with the method that produced it and the date it was run, how to read one covers what each field does and does not cover. ACE-031 (ramatercept) is a FUSION PROTEIN: the extracellular domain of activin receptor type-2B joined to an antibody Fc region. UniProt Q13705 is the receptor it derives from, not the fusion, which is a larger glycosylated protein whose mass the lot certificate reports. So the mass on the lot certificate is the one to read; a listing that quotes the published figure for this material is quoting the wrong molecule.

ACE-031 research: what the published studies examine

Status: No FDA-approved application. This compound is sold as laboratory material. Where research on it has been published, it sits at the animal, cell or early-human stage, and this record does not summarize what it found. A supplier is not a neutral judge of evidence about its own catalog.

ACE-031 is not a peptide but a fusion protein: the extracellular ligand-binding domain of activin receptor type IIB joined to an IgG1 Fc. It works as a decoy, mopping up myostatin and related ligands before they can signal. That mechanism is deliberately broad — activin type II receptors bind several TGF-beta superfamily members, not myostatin alone — and the breadth is the likely explanation for what happened in the clinic. The compound produced effects outside muscle, and those effects, rather than any lack of muscle activity, ended the program.

ACE-031: reported findings and the model each came from
Reported observationModelSource
In a randomised, double-blind, placebo-controlled ascending-dose trial in ambulatory boys with Duchenne muscular dystrophy, the study was STOPPED after the second dosing regimen over potential safety concerns of epistaxis and telangiectasias. Trends were reported for maintenance of six-minute walk distance against a decline in placebo, and for increased lean body mass and bone mineral density with reduced fat mass — but the walk-test trend was not statistically significant. The authors record that non-muscle-related adverse events contributed to the decision to discontinue.randomised double-blind placebo-controlled ascending-dose trial, ambulatory boys with Duchenne muscular dystrophyCampbell and colleagues, Myostatin inhibitor ACE-031 treatment of ambulatory boys with Duchenne muscular dystrophy: results of a randomized, placebo-controlled clinical trial, Muscle and Nerve (2016)Sponsor co-authored with academic investigators. The trial-stopping events are the material finding, not the efficacy trends.
The target rationale rests on myostatin being a negative regulator of skeletal muscle growth: mice with the GDF-8 gene disrupted show a widespread increase in muscle mass through hyperplasia and hypertrophy.gene targeting in miceMcPherron, Lawler and Lee, Regulation of skeletal muscle mass in mice by a new TGF-beta superfamily member, Nature (1997)

Open questions. The defining fact about ACE-031 is that its clinical trial was halted early over safety, specifically nosebleeds and telangiectasias — small dilated vessels visible in the skin. Those are vascular findings, not muscle findings, and they are consistent with the decoy binding TGF-beta superfamily ligands beyond myostatin. Any account of this compound that reports the lean-mass and bone-density trends without the discontinuation is describing half of a published result. The efficacy signals themselves were trends and the walk-test difference did not reach significance.

There is no approved product, and the program did not continue. Note also that this is a large Fc fusion protein rather than a peptide, which has consequences for handling, stability and what an identity assay on a lot would even involve.

Each row above describes an observation in the stated model. None of it describes what ACE-031 would do in a person or an animal outside that model, and none of it is a recommendation to use it for anything.

How to store ACE-031: refrigeration and shelf life

Keep the sealed vial cold and out of the light and it stays as it left the lab. Once it is mixed into a solution, how long it holds up depends on the compound, what it was mixed with, how strong the solution is and how cold it is kept — too many variables for anyone to print one shelf life on a label. So laboratories write the date on whatever they prepare and keep their own record of it.

For solution preparation arithmetic on the bench, concentration, per-graduation values and molarity, use the vial concentration calculator, which opens with the ACE-031 sizes stocked here already loaded. It covers laboratory measurement only and contains no administration guidance.
